Scientific Research Findings: Unveiling the Secrets of Female Anti-Aging

Poor ovarian health leads to premature aging...

As a crucial reproductive organ in females,the ovary plays a vital role in women's health.However,modern women's habits such as staying up late,prolonged sitting,excessive dieting,and binge eating have led to a trend of ovarian aging occurring at younger ages.

Irregular menstruation,acne,weight gain,poor sleep...These seemingly unrelated symptoms all point to issues like premature ovarian failure in women.

Premature Ovarian Failure(POF)

Premature ovarian failure(POF)refers to the phenomenon of amenorrhea occurring in women before the age of 40 due to ovarian function decline.Amenorrhea is accompanied by elevated levels of gonadotropins and decreased estrogen levels,along with varying degrees of hypoestrogenic symptoms such as hot flashes,sweating,facial flushing,and low libido.This disease can affect fertility and is hereditary.Premature ovarian failure caused by well-defined autoimmune diseases can achieve definite effects through immunosuppressive therapy.

Hydrogen-Oxygen Care for Ovarian Health

Considering the anti-inflammatory and antioxidant mechanisms of hydrogen-oxygen have the potential to improve the cellular microenvironment and reverse the aging process,scientists from Tianjin Medical University General Hospital studied the effects of hydrogen-oxygen on women's health and published a related SCI paper in the international journal Peer J.The paper elucidates the mechanism of hydrogen molecules in premature ovarian failure,indicating that hydrogen molecules can have a protective effect on the ovaries,significantly reducing ovarian damage caused by POF(premature ovarian failure),and greatly mitigating the health impacts of external stress-induced premature aging in modern women.[1]

Experimental Design

In this experiment,the scientists selected adult female rats as the animal model and established three groups:the control group,the POF group,and the POF+H2 group(POF stands for"Premature Ovarian Failure").

—Because cyclophosphamide(CTX)can induce premature ovarian failure in female rats,the POF group and the POF+H2 group in this experiment were injected with cyclophosphamide,while the control group was injected with an equal volume of saline.The POF+H2 group was then allowed to inhale hydrogen molecules.Finally,the differences in various indicators among the three groups were observed to compare the impact of hydrogen molecules on POF.(These indicators are all related to the severity of premature ovarian failure.Comparing these indicators can provide an intuitive understanding of the severity of premature ovarian failure.)

Experimental Results

1.Indicator Testing

Scientists tested multiple indicators across three groups.The indicators in the figures below are:A for"Estradiol(E2)",B for"Follicle-Stimulating Hormone(FSH)",C for"Anti-Müllerian Hormone(AMH)",and D for"Follicle Numbers".

Group A:Estradiol(E2):Estradiol is a crucial indicator in gonadal hormone testing.It is the most active and physiologically significant component of estrogen.Elevated estradiol levels are commonly observed in pregnant women and elderly men,while decreased estradiol levels indicate conditions such as premature ovarian failure.

Group B:Follicle-Stimulating Hormone(FSH):Follicle-Stimulating Hormone,also known as FSH,is an important gonadotropin.FSH levels increase with age,so elevated FSH indicates declining ovarian function,and conditions like premature ovarian failure and amenorrhea can lead to increased FSH levels.

Group C:Anti-Müllerian Hormone(AMH):It can be used to assess ovarian reserve function and predict the timing of menopause in women.Higher AMH levels indicate a greater egg reserve,while lower AMH levels indicate poorer ovarian function.

Group D:Measurement of follicle counts at various stages.Comparing the POF,POF+H2 groups with the control group,a larger difference from the control group indicates poorer ovarian function.


As shown in the figure above,Premature Ovarian Failure(POF)leads to decreased estradiol(E2),increased follicle-stimulating hormone(FSH),increased Anti-Müllerian Hormone(AMH),and reduced follicle counts at various stages.In contrast,hydrogen molecule intake can significantly mitigate the effects of POF and improve ovarian health.

2.Morphological Observation

Scientists observed the follicle morphology in the three groups.In the figure below,"PF"stands for"Primordial Follicle,""SF"stands for"Secondary Follicle,"and"AF"stands for"Antral Follicle"(Antral follicles are mature follicles).The findings are as follows:


As shown in the figure above,after hydrogen molecule intake,the POF+H2 group exhibited a greater number of follicles in better condition.

The figure below uses fluorescent staining to highlight apoptotic cells for calculating the proportion of apoptotic cells.The results show that hydrogen molecule administration reduced the number of apoptotic cells,thereby protecting the oocytes.


3.Protein Expression

To investigate the reasons why hydrogen affects ovarian cells,scientists compared the differences in protein expression in ovarian cells.The results showed that in the POF group,there were 48 upregulated proteins and 58 downregulated proteins,while in the POF+H2 group,there were 27 upregulated proteins and 58 downregulated proteins.This indicates that hydrogen intake is involved in the protein expression within the ovaries,repairing the impact of POF on protein expression.The results are shown in the figure below:


4.Gene Targets

Protein expression is determined by genes.If the protein expression in POF shows differences due to hydrogen molecule uptake,it indicates that hydrogen acts on certain gene targets,reducing the differences in protein expression and thereby protecting the ovaries.Through methods such as GO(Gene Ontology)functional enrichment analysis and KEGG(Kyoto Encyclopedia of Genes and Genomes)analysis,scientists identified a total of 16 candidate differentially expressed proteins(DEPs).These proteins were found to be significantly enriched in 296 GO terms and 36 KEGG pathways.

Based on protein-protein interaction networks and the GeneMANIA network,the key targets RT1-Db1 and RT1-Bb were ultimately identified.

Results and Discussion

RT1-Db1andRT1-Bbare homologous to human HLA-DRB5 and HLA-DQB1,respectively.Both HLA-DRB5 and HLA-DQB1 belong to MHC class II proteins[2].It is known that the deficiency of the MHC class II accessory molecule HLA-DO is associated with susceptibility to autoimmune diseases.In this study,the expression of RT1-Db1 and RT1-Bb was significantly reduced in POF,but markedly improved after hydrogen molecule inhalation,suggesting that hydrogen gas may influence the expression of class II proteins.Therefore,hydrogen may alleviate ovarian damage in rats with premature ovarian failure by targeting class II proteins.

Due to its anti-inflammatory and antioxidant mechanisms,as well as its characteristic of gently intervening in signal transduction,hydrogen-oxygen inhalation has been widely applied in the field of women's health research.Scientists from the Naval Medical University(Second Military Medical University)used high-concentration hydrogen-oxygen mixed inhalation(66.6%hydrogen+33.3%oxygen)to improve endometriosis.They found that high-concentration hydrogen-oxygen inhalation can significantly reduce the oxidative stress index in endometrial tissue and decrease the expression of MMP-9 and COX-2,thereby alleviating endometriosis[3].

Furthermore,in related research conducted at Beijing University of Technology,scientists discovered that high-concentration hydrogen-oxygen mixed inhalation(66.6%hydrogen+33.3%oxygen)can inhibit the growth of ovarian cancer.Hydrogen-oxygen inhalation can suppress the proliferation of cancer stem-like cells(CSCs)and angiogenesis,which is considered beneficial for anticancer effects[4].

The Revelation of Hydrogen-Oxygen

"Daughters are made of water..."

This line,spoken by Jia Baoyu in Dream of the Red Chamber by author Cao Xueqin,praises women for being as gentle and pure as water.During the clinical application of hydrogen-oxygen medicine,many women have reported that their skin regained its luster and elasticity,and their overall health improved.This brings to mind that hydrogen-oxygen mixed gas(66.6%hydrogen+33.3%oxygen)is derived from natural water molecules...Using water to improve women's health seems to align with the profound principles of nature's way.
